From Adobe Kuler there are losts of nice color schema's available but in general what should one do; A dark brackground and lighter font or a light background and darkletter etc. I'm sure there is some research done but what in your experience is best?

Personally, I think that unless you have good reason, stick with darker text on a lighter back ground (white or light grey).
I usually use very dark grey text rather than black, so maybe #333333. It's just a bit softer on the eye.
That said, there are times when light text on a dark background is good too, but only when there's not too much text to read. A dark background can look very elegant and convey a feel of of expense, eg:
